Convertible Solid Supports for 3'-Modification of Oligonucleotides
Convertible solid supports utilize the exceptional selectivity of the thioester function towards primary amines and are used for the introduction of ligands and functional groups at the 3'-terminus of synthetic oligonucleotides. Examples of the successful derivatization include 3'- aminoalkyl, mercaptoalkyl, aldehydo, hydrazido, carboxy, imidazole, TEMPO, PROXYL, and lipophylic alkyl groups. Our experience with these solid supports exceeds 10 years, which allowed us to develop a set of very reliable and user-friendly protocols available in pdf format.
